The Distribution of Histochemical Fiber Types in the Tibialis Anterior and Soleus Muscles of the Adult Rat


Abstract
It is well known that the muscle fibers of the mammalian skeletal muscle can be differentiated histochemically by the intensity of staining reaction .and enzyme activities in the sarcoplasms, which are best characterized on flash frozen sections. Mammalian skeletal muscles possess a heterogeneous population of histochemical fiber types which are arranged in a mosaic pattern.
Yet few reports have described that proportion of each fiber type varies among individual muscles within an animal, and homologous muscles of different species.
This experiment was designed for the evaluation of differences in distri.bution of the fibers according to anatomical region classified by the superficial to deep and medial to lateral aspects of the tibialis anterior and the soleus muscles in the adult rats. Trichrome, H-E, PAS, myosin-ATPase and succinate dehydrogenase (SDH) stains were performed in transverse sections of the muscle.
The main results of this study were as follows.
1. The tibialis anterior muscle was composed of three fiber types (type I, type IIA and type IIB) which can be differentiated by staining for myosin ATPase and SDH. On the other hand, the soleus muscle had only two fiber types with absence of type IIB fiber.
2. The soleus muscle had a greater proportion of the type I fibers in comparison with the tibialis anterior muscle. But, PAS staining revealed no difference in distribution of fiber types, between the tibialis anterior and soleus muscles.
3. Type IIA fibers, which showed high levels of myosin-ATPase and SDH activities, were more abundant in the deep region of the tibialis anterior and soleus muscles. On the contrary, type IIB fibers in the tibialis anterior were predominant in the superficial and lateral regions.
4. Periodic acid-Schiff positive fiberss showed more or less even distribution regardless of the region in the tibialis anterior and soleus muscles.
